Este avance podr\u00eda facilitar el desarrollo de terapias m\u00e1s precisas para cada paciente y refuerza la idea de que el trastorno no constituye una condici\u00f3n \u00fanica con una sola causa biol\u00f3gica.<\/p>\n<p>Investigaciones anteriores hab\u00edan intentado detectar patrones comunes para clasificar diferentes formas de autismo mediante el an\u00e1lisis de cientos de rasgos conductuales o el <a href=\"https:\/\/es.wired.com\/articulos\/autismo-puede-ser-entendido-con-organoides-creados-en-laboratorios\">estudio de la relaci\u00f3n entre la gen\u00e9tica<\/a> y la etapa de la vida en la que se manifiesta. Estudio revela que quienes son diagnosticados temprano y quienes lo son m\u00e1s tarde muestran perfiles gen\u00e9ticos y de desarrollo distintos, lo que redefine su comprensi\u00f3n y tratamiento.<\/p>\n<\/div>\n<\/div>\n<hr>\n<p>Alessandro Gozzi, investigador del Instituto Italiano de Tecnolog\u00eda y coautor principal del estudio, se\u00f1al\u00f3 que, aunque durante d\u00e9cadas se han observado variaciones en la manifestaci\u00f3n del autismo, hasta ahora exist\u00edan pocas pruebas de que esas diferencias reflejaran mecanismos biol\u00f3gicos subyacentes distintos. \u201cNuestro enfoque nos permiti\u00f3 aislar factores gen\u00e9ticos e inmunitarios espec\u00edficos y, posteriormente, trasladar esas caracter\u00edsticas a esc\u00e1neres cerebrales humanos, demostrando que distintos patrones de conectividad codifican diferentes v\u00edas mecan\u00edsticas relacionadas con el autismo\u201d, <a href=\"https:\/\/www.eurekalert.org\/news-releases\/1130160\">explic\u00f3<\/a> el especialista.<\/p>\n<p>Gozzi y sus colegas analizaron im\u00e1genes cerebrales obtenidas por resonancia magn\u00e9tica funcional de 20 modelos gen\u00e9ticos distintos de ratones que presentaban caracter\u00edsticas asociadas con el autismo. Esta t\u00e9cnica permiti\u00f3 observar c\u00f3mo se comunican entre s\u00ed las diferentes regiones del cerebro.<\/p>\n<p>A partir de estos datos, <strong>los investigadores identificaron alteraciones en la conectividad cerebral que pod\u00edan agruparse en dos categor\u00edas principales.<\/strong> En la primera, las conexiones eran m\u00e1s d\u00e9biles de lo habitual, un fen\u00f3meno conocido como hipoconectividad. En la segunda ocurr\u00eda lo contrario: la comunicaci\u00f3n entre las distintas \u00e1reas cerebrales era m\u00e1s intensa de lo normal, lo que se denomina hiperconectividad.<\/p>\n<p>Los resultados iniciales mostraron que ambos patrones estaban relacionados con mecanismos biol\u00f3gicos diferentes. La hipoconectividad parec\u00eda asociarse con alteraciones en las sinapsis, es decir, en los puntos de comunicaci\u00f3n entre las neuronas. Por el contrario, la hiperconectividad estaba vinculada con cambios en la actividad de determinados genes y con modificaciones en procesos del sistema inmunitario.<\/p>\n<p>Adriana Di Martino, cient\u00edfica del Child Mind Institute y tambi\u00e9n coautora principal del estudio, afirm\u00f3 que el trabajo realizado con ratones constituy\u00f3 la base para comprender mejor la biolog\u00eda cerebral del autismo. \u201cPudimos observar qu\u00e9 v\u00edas biol\u00f3gicas impulsan determinados patrones de conectividad y, posteriormente, buscar esas mismas se\u00f1ales en humanos\u201d, indic\u00f3.<\/p>\n<h2>De modelos murinos a pacientes humanos<\/h2>\n<p>En una segunda fase, los cient\u00edficos intentaron determinar si estos patrones pod\u00edan reproducirse en personas. Para ello, analizaron im\u00e1genes cerebrales de casi 2,000 individuos procedentes de diversos centros de investigaci\u00f3n y bases de datos. La muestra incluy\u00f3 a 940 personas con autismo y a m\u00e1s de 1,000 participantes sin esta condici\u00f3n.<\/p>\n<p>La evaluaci\u00f3n revel\u00f3 que los patrones de hipoconectividad e hiperconectividad tambi\u00e9n estaban presentes en los cerebros humanos, al igual que los mecanismos biol\u00f3gicos detectados en los modelos animales. En particular, se mantuvieron las asociaciones con alteraciones sin\u00e1pticas y con procesos inmunitarios y gen\u00e9ticos. En conjunto, <strong>estos dos subtipos representaron aproximadamente el 25% de las personas con autismo incluidas en el estudio.<\/strong><\/p>\n<p>Adem\u00e1s de las diferencias observadas en la funci\u00f3n cerebral, los investigadores identificaron <a href=\"https:\/\/es.wired.com\/articulos\/inducen-autismo-en-perros-para-estudiar-este-trastorno\">caracter\u00edsticas conductuales<\/a> particulares y una organizaci\u00f3n espec\u00edfica de las redes neuronales en cada uno de los grupos.<\/p>\n<p>Los autores sostienen que la detecci\u00f3n de los mismos subtipos en decenas de centros de investigaci\u00f3n independientes constituye una validaci\u00f3n fundamental de sus conclusiones.
